A panel of highly experienced investigators describe in step-by-step fashion key techniques for experimentally detecting SV40 in human tumors, for exploiting its use in human gene therapy, and for studying its replication and its mechanisms of neoplastic transformation. Included are methods for growing SV40 and related viruses in tissue culture, for in vivo and in vitro replication and transcription of SV40 DNA, for the use of retroviral vectors to express SV40 tumor antigens in cultured cells, and the use of transgenic mouse models based on the SV40 large T antigen. Detailed and highly practical, SV40 Protocols offers both clinical and basic researchers powerful, well-tested tools for research on SV40 replication and neoplastic transformation, as well as techniques for its detection in human tumors and for creating and using powerful new gene therapy vectors.

Reviewer:Jerry Lynn Taylor, PhD(Medical College of Wisconsin)
Description:This book contains a collection of basic procedures that can be used to study a variety of aspects of SV40 virus and its effects on cells in culture and in animal models.
Purpose:This collection is designed to contain protocols that are currently used by workers with experience in SV40 research. Because descriptions of experimental design in the primary literature are often incomplete, this grouping of protocols was created to help researchers more easily study and use SV40 virus.
Audience:This book is targeted at both beginners as well as experienced researchers in virology, cell biology, oncology, and immunology providing detailed protocols for studying the virus as well as using the virus and its proteins as tools to explore cellular activities.
Features:The protocols included describe in specific detail how studies are done, how they are interpreted, what potential problems may arise, and the solutions to problems. In addition to a useful index, there is a list of commercial suppliers whose products are cited in the text. Contact numbers including Internet addresses for these suppliers are included. Black-and-white figures and tables provide useful information and illustrate experimental design as well as potential results.
Assessment:This should be an extremely useful practical guide for those planning to work with the SV40 virus or use SV40 T antigens as tools to study biological processes.
